Various characteristics between the two groups cause a bacteria to be either gram-positive or gram-negative, but one of the most significant traits that distinguish a gram-positive bacteria from a gram-negative bacteria is the difference of peptidoglycan in their cell wall. Gram-positive bacteria have a rather thick layer of peptidoglycan, whereas a gram-negative bacteria has a thin layer of peptidoglycan followed by an outer membrane (Smith & Hussey).
Another stain important in the field of microbiology is the endospore stain. Endospores are dormant cells that are highly resistant to heat and other harmful factors (Allen, Anderson & Salm, 2016). Due to this reason, endospores require a different technique called the endospore stain, which is used to make endospores easier to see under a microscope (Hussey & Zayaitz). This stain requires a steamer due to the fact that endospores are highly resistant to heat.
